BH3-Only Molecule Bim Mediates ß-Cell Death in IRS2 Deficiency
Irs2 deficient mice develop type2- like diabetes due to a reduction in ß-cell mass and a failure of pancreatic islets to undergo compensatory hyperplasia in response to insulin resistance. In order to define the molecular mechanisms, researchers knocked down Irs2 gene expression in mouse MIN6 insulinoma cells. IRS2 suppression induced apoptotic cell death which was associated with an increase in expression of the BH3-only molecule Bim. [Diabetes] Abstract

IL-6-Dependent Proliferation of Alpha Cells in Mice with Partial Pancreatic-Duct Ligation
As beta cells expand following partial pancreatic-duct ligation (PDL) in adult mice, scientists investigated whether PDL also causes alpha cells to expand and whether IL-6 signaling is involved. [Diabetologia] Abstract

Inefficient Translocation of Preproinsulin Contributes to Pancreatic Beta Cell Failure and Late Onset Diabetes
Researchers provide the first evidence that inefficient translocation of preproinsulin contributes to beta cell failure and diabetes. Specifically, they found that after targeting to the endoplasmic reticulum (ER) membrane, preproinsulin signal peptide mutants associated with autosomal dominant late-onset diabetes fail to be fully translocated across the ER membrane. Distinct Cell Clusters Touching Islet Cells Induce Islet Cell Replication in Association with Over-Expression of Regenerating Gene (REG) Protein in Fulminant Type 1 Diabetes
The architectures around islet cell clusters, including basement membranes, extracellular matrix, and pancreatic acinar-like cell clusters, were studied in the non-diabetic state and in the inflamed milieu of fulminant type 1 diabetes in humans. [PLoS One] Full Article

Quantitative-Proteomic Comparison of Alpha and Beta Cells to Uncover Novel Targets for Lineage Reprogramming
Scientists compared the transcriptome, proteome, and phosphoproteome of alpha cells, beta cells, and compound-treated alpha cells. Their phosphoproteomic analysis indicated that two kinases, BRSK1 and CAMKK2, exhibit decreased phosphorylation in beta cells compared to alpha cells, and in compound-treated alpha cells compared to DMSO-treated alpha cells. miR-615-5p Is Epigenetically Inactivated and Functions as a Tumor Suppressor in Pancreatic Ductal Adenocarcinoma
Using methylated DNA immunoprecipitation-chip analysis, scientists found that microRNA (miR)-615-5p was hypermethylated in its putative promoter region, which silenced its expression in pancreatic ductal adenocarcinoma cell lines. In addition, the overexpression of miR-615-5p in pancreatic cancer cells suppressed cell proliferation, migration and invasion. [Oncogene] Abstract

APPL Proteins Modulate DNA Repair and Radiation Survival of Pancreatic Carcinoma Cells by Regulating ATM
Researchers showed that adaptor proteins containing pleckstrin homology domain, phosphotyrosine binding domain and a leucine zipper motif (APPL)2 expression in pancreatic cancer cells is upregulated after irradiation and that depletion of APPL proteins by small interfering RNA significantly reduced radiation survival in parallel to impairing DNA double strand break repair. Antitumor Activities of Rauwolfia vomitoria Extract and Potentiation of Gemcitabine Effects against Pancreatic Cancer
A ß-carboline-enriched extract from the plant Rauwolfia vomitoria induced apoptosis in pancreatic cancer cells in a concentration-dependent manner, and completely inhibited colony formation of PANC-1 cells in soft agar. [Integr Cancer Ther] Abstract

Polyserase-1/TMPRSS9 Induces Pro-Tumor Effects in Pancreatic Cancer Cells by Activation of Pro-uPA
Researchers evaluated the possible role of polyserase-1 in cancer progression. They showed that polyserase-1 increased the invasive capacities of PANC-1 and SK-PC-3 pancreatic cancer cells. [Oncol Rep] Abstract

Novel Drug Cocktail May Improve Clinical Treatment for Pancreatic Cancer
Scientists used pancreatic cancer cells to assess the efficacy of combining doxorubicin (DOX) with a drug that inhibits the survival proteins involved in DOX resistance called BEZ235 (BEZ). The results showed that treatment with the combination of DOX and BEZ had significantly lower rate of surviving pancreatic cancer cells than the cells with single drug treatment. Pancreatic Tumor-Induced Gene May Prove Beneficial as a Drug Marker
Scientists were able to represent images of pancreatic Rgs16 expression, and hence all the stages of pancreatic tumor development, quantitatively. This helped them not only lay out the progression of the disease explicitly, but also opened up a new avenue: testing drug effects on tumor growth.
