Researchers Identify New Drug Target That Stimulates Beta Cell Regeneration
One of the holy grails in diabetes research is to discover molecules that stimulate beta cell growth and to find drugs that target these molecules. Now, researchers have done both, discovering not only a protein that regulates beta cell growth, but also a chemical compound that stimulates it. [Press release from the Juvenile Diabetes Research Foundation discussing online prepublication in Cell Metabolism] Press Release | Abstract

DIABETES

Pancreatic Mesenchyme Regulates Epithelial Organogenesis Throughout Development
The results demonstrate that mesenchymal cells regulate pancreatic growth and branching at both early and late developmental stages by supporting proliferation of precursors and differentiated cells, respectively. [PLoS Biol] Abstract | Press Release

Expansion of Th17 Cells and Functional Defects in T Regulatory Cells Are Key Features of the Pancreatic Lymph Nodes in Patients With Type 1 Diabetes
The data provides evidence for an unbalanced immune status in the pancreatic lymph nodes of type 1 diabetic subjects, and treatments restoring the immune homeostasis in the target organ of these patients represent a potential therapeutic strategy. [Diabetes] Abstract

Mechanisms of Control of the Free Ca2+ Concentration in the Endoplasmic Reticulum of Mouse Pancreatic β-Cells Interplay With Cell Metabolism and [Ca2+], and Role of SERCA2b and SERCA3
Sarco-endoplasmic reticulum Ca2+-ATPase 2b (SERCA2b) and SERCA3 pump Ca2+ in the endoplasmic reticulum (ER) of pancreatic β-cells. Investigators studied their role in the control of the free ER Ca2+ concentration and the role of SERCA3 in the control of insulin secretion and ER stress. [Diabetes] Abstract

Perfluorodecalin-Enriched Fibrin Matrix for Human Islet Culture
The study aimed to create a matrix for culturing islets, using fibrin as scaffold and perfluorodecalin as oxygen diffusion enhancing medium. [Biomaterials] Abstract

A Novel Explant Outgrowth Culture Model for Mouse Pancreatic Acinar Cells with Long-Term Maintenance of Secretory Phenotype
Despite considerable efforts, no long-term culture models for non-transformed pancreatic acini exist. The aim was to develop and validate culture conditions for this purpose. [Eur J Cell Biol] Abstract

PANCREATIC CANCER

Pancreatic Ductal and Acinar Cell Neoplasms in Carney Complex: A Possible New Association
Immunohistochemistry and loss-of-heterozygosity studies suggest that PRKAR1A could function as a tumor suppressor gene in pancreatic tissue, at least in the context of Carney complex. [J Clin Endocrinol Metab] Abstract

Cationic Liposomal Paclitaxel Plus Gemcitabine or Gemcitabine Alone in Patients with Advanced Pancreatic Cancer: A Randomized Controlled Phase II Trial
Paclitaxel embedded in cationic liposomes (EndoTAG™-1; ET) is an innovative agent targeting tumor endothelial cells. This randomized controlled phase II trial evaluated the safety and efficacy of ET in combination with gemcitabine in advanced pancreatic cancer. [Ann Oncol] Abstract

Inhibition of Focal Adhesion Kinase by PF-562,271 Inhibits the Growth and Metastasis of Pancreatic Cancer Concomitant with Altering the Tumor Microenvironment
Researchers investigated the effects of PF-562,271 (a small molecule inhibitor of FAK/PYK2) on in vitro migration, invasion and proliferation, tumor proliferation, invasion and metastasis in a murine model, and stromal cell composition in the pancreatic ductal adenocarcinoma microenvironment. [Mol Cancer Ther] Abstract

Targeting Epigenetic Regulation of miR-34a for Treatment of Pancreatic Cancer by Inhibition of Pancreatic Cancer Stem Cells
The study aimed to investigate the functional significance of microRNA (miR)-34a in pancreatic cancer progression through its epigenetic restoration with chromatin modulators, demethylating agent 5-Aza-2′-deoxycytidine and HDAC inhibitor Vorinostat (SAHA). [PLoS One] Abstract

Proteomic Analysis of Gemcitabine-Induced Drug Resistance in Pancreatic Cancer Cells
To investigate the molecular mechanisms associated with gemcitabine (GEM)-induced resistance, 2D-DIGE and MALDI-TOF mass spectrometry were performed to compare the proteomic alterations of a panel of differential GEM-resistant PANC-1 cells with GEM-sensitive pancreatic cells. [Mol Biosyst] Abstract
